Japanese Encephalitis Virus (JEV)

Structure and Key Research Targets

Japanese Encephalitis Virus (JEV) structure

Overview

Japanese encephalitis virus (JEV) is an enveloped, positive-sense single-stranded RNA virus belonging to the genus Flavivirus within the family Flaviviridae. JEV infection primarily affects the central nervous system and is an important model for studying flavivirus structure, viral replication, immune recognition, and antibody responses. The Envelope protein (E) is a major surface antigen involved in viral entry and neutralizing antibody recognition, making it a key target for JEV research.

JEV Structure

JEV is an approximately 40-50 nm enveloped virus containing an approximately 11 kb positive-sense single-stranded RNA genome. The viral surface is mainly composed of E and M proteins arranged on the lipid envelope. The E protein forms the outer protein shell and plays essential roles in receptor binding and membrane fusion during viral entry. The internal nucleocapsid contains viral RNA associated with the capsid protein, while non-structural proteins participate in viral RNA replication and polyprotein processing.

The viral genome encodes a single polyprotein that is processed into three structural proteins and seven non-structural proteins:

  • Structural proteins: Capsid (C) protein, Premembrane/Membrane (prM/M) protein, Envelope (E) protein
  • Non-structural proteins: NS1, NS2A, NS2B, NS3, NS4A, NS4B, NS5

Key Research Targets

Envelope Protein (E): Major Neutralizing Antigen Key Target
The Envelope protein (E) is the most extensively studied JEV structural protein. E protein contains three major domains: Domain I (EDI) central structural domain, Domain II (EDII) containing the fusion loop responsible for membrane fusion, and Domain III (EDIII) involved in receptor interaction and containing important neutralizing antibody epitopes. During infection, E protein mediates viral attachment to host cells, membrane fusion, viral entry, and recognition by neutralizing antibodies. Because of its exposed location on the viral surface and immunogenic properties, JEV E protein is a major target for antibody development, neutralization assays, serological detection, and vaccine-related research.
Applications: Antibody development · Neutralization assays · Serological detection · Vaccine research
Neutralization Vaccine Entry Antibody
prM/M Protein: Viral Maturation and Assembly Maturation
The prM/M protein plays an important role in JEV particle maturation. During viral assembly, prM associates with E protein and stabilizes immature viral particles. Following cleavage of prM during maturation, mature M protein remains associated with the viral envelope and contributes to formation of infectious virions. Research on prM/M supports studies of viral assembly, virion maturation, and flavivirus structural biology.
Applications: Viral assembly · Virion maturation · Flavivirus structural biology

NS1 Protein: Viral Replication and Immune Response Immune Target
NS1 is a conserved non-structural protein involved in the formation of viral replication complexes. JEV NS1 research focuses on viral replication mechanisms, host-virus interactions, antigen detection, and immune response analysis. Because NS1 can be secreted during flavivirus infection, it is also investigated as a potential diagnostic and immune-related target.
Applications: Viral replication mechanisms · Host-virus interactions · Antigen detection · Immune response analysis
Replication Antigen detection Immunology
NS3 and NS5 Proteins: Viral Replication Machinery Replication
The non-structural proteins NS3 and NS5 are essential components of JEV replication. NS3 contains protease and helicase activities involved in viral polyprotein processing and RNA replication. NS5 functions as the viral RNA-dependent RNA polymerase and methyltransferase. These proteins are important targets for molecular virology studies, antiviral research, and viral replication mechanism analysis.
Applications: Molecular virology studies · Antiviral research · Viral replication mechanism analysis
